Analysis of in vivo Mutation at hprt Gene from the Rat Exposed to Pentachlorophenol

Abstract
The mutagenic potential of pentachlorophenol (PCP) was measured at the hprt(hypoxanthine guanine phosphoribosyl transferase) locus in blood and splenic T-cells from rats in vivo. Rats were grown for 1, 6 and 12 months under optional condition fed
with
food supplements with PCP (0~2,000ppm, weight/weight). Spleens and blood were isolated from these animals and both cell types were stimulated with concanavalin A and interleukin 2 and then seeded in medium containing 6-thioguanine to select for
hprt
negative mutants.
Pentachlorophenol produced statistically significant increase in the mutant fraction except at high dose of PCP (742mg net dose at 12 month). The relationship between the net dose of PCP and the mutant fraction in hprt gene was approximately
linear
over
the range of 0~400mg of PCP with an linear regression equation of [mutant fraction={8.89x(net of PCP in mg)+1.967}x10E-10; in splenic cell] and [mutant fraction={10.40x(net dose of PCP in mg)+1.472}x10E-10; in blood cell].
